Daptomycin

Daptomycin, being a cyclic lipopeptide, it belongs to a new class of antibiotics. It acts by formation of a pore in the bacterial membrane (1), causing leakage of potassium (2) and subsequent depolarization (3). This results in an arrest of bacterial cell function (DNA, RNA and protein synthesis; 4) and finally cell death (5).

Daptomycin has a bactericidal action on Gram-positive bacteria and is now registered for treatment of complicated skin and soft tissue infections caused by Gram-positive microorganisms in adults. There is limited experience with treatment of S. aureus bacteriemia.

The most important adverse effects include gastrointestinal complaints, skin reactions at the infusion site and increased serum creatinefosfokinase.
