Ketoconazole

Ketoconazole inhibits two enzymes in the glucocorticoid synthesis. It blocks the cholesterol side chain cleavage enzyme, which converts cholesterol into pregnenolone and it inhibits the 11β-hydroxylase enzyme (like metyrapone).

The final effect of ketoconazole is a decreased glucocorticoid synthesis. This drug can be used to control adrenal hyperfunction, caused by Cushing's syndrome or Cushing's disease. Remember the role of ketoconazole in infectious diseases.
